چکیده

The electric field method of gene transfer via electroporation of cell membranes is gaining importance in cellular and molecular biology. In particular the electroporation technique provides the low copy number transfection required fqr genetic studies. As demonstrated with mouse lyoma cells, with lymphoid cell lines and with plant protoplast, the electric method has appreciable advantages compared to biochemical procedures of cell transformation. Electroporation also provides a physically plausible mechanism for cell fusion in electric pulses.

Transfection of bovine spermatogonial stem cells in vitro

Spermatogonial stem cells (SSCs) are the only stem cells in adults that can transfer genetic information to the future generations. Considering the fact that a single SSC gives rise to a vast number of spermatozoa, genetic manipulation of these cells is a potential novel technology with feasible application to various animal species. The aim of this study was to evaluate enhanced green fluoresc...

The Effect of Wild Type P53 Gene Transfer on Growth Properties and Tumorigenicity of PANC-1 Tumor Cell Line

The p53 protein function is essential for the maintenance of the nontumorigenic cell phenotype. Pancreatic tumor cells show a very high frequency of p53 mutation. To determine if restoration of wild type p53 function can be used to eliminate the tumorigenic phenotype in these cells, pancreatic tumor cell lines, PANC-1 and HTB80, differing in p53 status were stably transfected with exogenous wil...

Electric field mediated gene transfer.

A simple physical chemical method of transferring genes into eucaryotic cells is described. Electric impulses in the intensity range of 5-10 kV/cm with a duration of 5-10 bs were found to appreciably increase the uptake of DNA into cells. After electric field treatment, stable transformants were obtained in a system containing mouse cells deficient in thymidine kinase and a plasmid DNA harbouri...

Gene transfer into mouse lyoma cells by electroporation in high electric fields.

Electric impulses (8 kV/cm, 5 microseconds) were found to increase greatly the uptake of DNA into cells. When linear or circular plasmid DNA containing the herpes simplex thymidine kinase (TK) gene is added to a suspension of mouse L cells deficient in the TK gene and the cells are then exposed to electric fields, stable transformants are formed that survive in the HAT selection medium. At 20 d...

The effect of high frequency electric field on enhancement of chondrogenesis in human adipose-derived stem cells

Objective(s):Osteoarthritis (OA) is globally one of the most common diseases from the middle age onwards. Cartilage is an avascular tissue therefore it cannot be repaired in the body. Conservative treatments have failed as a good remedy and cell therapy as a decisive cure is needed. One of the best and easily accessible cell sources for this purpose is adipose-derived stem cells which can be di...
